RaduAricescu<sup>13<\/sup>ReneAssenberg<sup>2<\/sup>Robin T.Aplin<sup>2<\/sup>Robert J.C.Gilbert<sup>13<\/sup>Jonathan M.Grimes<sup>123<\/sup>David I.Stuart<sup>123<\/sup><\/a><\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Summary<\/h2>\n\n\n\n<p style=\"font-size:24px\">To achieve the greatest output from their limited\u00a0<a href=\"https:\/\/www.sciencedirect.com\/topics\/biochemistry-genetics-and-molecular-biology\/virus-genome\">genomes, viruses<\/a>\u00a0frequently make use of alternative\u00a0<a href=\"https:\/\/www.sciencedirect.com\/topics\/biochemistry-genetics-and-molecular-biology\/open-reading-frame\">open reading frames<\/a>, in which translation is initiated from a\u00a0<a href=\"https:\/\/www.sciencedirect.com\/topics\/biochemistry-genetics-and-molecular-biology\/start-codon\">start codon<\/a>\u00a0<em>within<\/em>\u00a0an existing gene and, being out of frame, gives rise to a distinct protein product. These alternative protein products are, as yet, poorly characterized structurally. Here we report the crystal structure of ORF-9b, an alternative open reading frame within the\u00a0<a href=\"https:\/\/www.sciencedirect.com\/topics\/biochemistry-genetics-and-molecular-biology\/virus-nucleocapsid\">nucleocapsid<\/a>\u00a0(N) gene from the\u00a0<a href=\"https:\/\/www.sciencedirect.com\/topics\/biochemistry-genetics-and-molecular-biology\/sars-coronavirus\">SARS coronavirus<\/a>. The protein has a novel fold, a dimeric tent-like \u03b2 structure with an amphipathic surface, and a central hydrophobic cavity that binds lipid molecules. This cavity is likely to be involved in membrane attachment and, in\u00a0<a href=\"https:\/\/www.sciencedirect.com\/topics\/biochemistry-genetics-and-molecular-biology\/mammalian-cell\">mammalian cells<\/a>, ORF-9b associates with intracellular vesicles, consistent with a role in the assembly of the virion.
